Emergency services responded to reports of a fire on Dorothy St in Fawkner after 2pm on Monday, with fire crews arriving to the house fully engulfed.
First responders commenced CPR on a child, aged three, but the child died at the scene.
Another adult occupant is being treated for non-life threatening injuries.
The cause of the fire is yet to be determined, a Victoria Police spokeswoman said.
Anyone who witnessed the incident, has dashcam or CCTV footage or anyone with information is urged to contact Crime Stoppers.
